DESCRIPTION

Melanin biosynthesis takes place in melanosomes, specialized melanocyte organelles, dendritic cells, located in the basal layer of the epidermis. L-Tyrosine and dopaquinone (derived from L-DOPA), are essential for their formation. L-Tyrosine and L-DOPA (L-dihydroxyphenylalanine) are, therefore, substrates for melanogenesis, and although they follow different metabolic pathways, they seem to be related with respect to the biochemical-physiological effects that lead to the formation of Melanin. In addition to being substrates for the formation of Melanin, L-Tyrosine and L-DOPA, act as positive inducers and regulators of melanogenic metabolic pathways, with direct or indirect interference from specific receptors and processes not mediated by receptors. The various melaninic components (eumelin and phaeomelanin) are related to the pigmentation of the hair follicle (follicular melanocytes, keratinocytes, dermal papilla fibroblasts). The capillary bulb, which contains a high amount of melanocytes, is essential for capillary pigmentation. L-Tyrosine and L-DOPA are also important in these biochemical-physiological processes (molecular physiology). The melanocytes of the black hair follicles contain the largest number of melanosomas with a high eumelanin content, which have a high electron density. The brown hair bulbs are more poorly melanized, having a preponderance of phaeomelanin, and in red hair the melanin is irregularly deposited in the form of flocculent material. With aging, melanocytes become dysfunctional and do not contain melanin, thus leading to hair whitening. Supplementation with melanin thus proves to be of enormous importance. L-Methionine is a sulfur amino acid that contributes to reversing the hair whitening process, as it prevents the oxidation of methionine itself and contributes to the formation of methionine sulfoxide reductase A and B, preventing the decrease in methionine's biochemical functionality. sulfoxide, thus contributing to the replacement of the dark color in the grayish follicle. As eumelanin is evident, it should counteract these biochemical-physiological deviations, and the use of L-Methionine as a synergist in the formulation, also plays a decisive role with regard to this activity. The addition of standardized extract of Piper nigrum to 95% piperine, to the formulation, aims to provide an increase in the bioavailability of the other components. Studies indicate that piperine increases assimilation at the intestinal level, but also contributes to increasing the time of metabolic availability of substances administered concomitantly, evidenced by the highest serum peaks and prolonged time of physiological activity. It is concluded that supplementation with Vitamin D is really highly relevant, since it lowers the risk of an older person falling by 22%.
